摘要(1)雄生殖系列特異的hsp70-2遺伝子の結合タンパク質TAF170は、そのプロモーター領域3カ所に結合し、発生時期特異的に雄生殖細胞に局在することを明らかにした。また、FKBP6はリボゾームの小サブユニットと相互作用すること、FKBP6欠損ラット生殖細胞では、集塊を形成したリボゾームを除くと野生型の30〜40%のリボゾームが細胞内に存在することを明らかにし、雄生殖細胞の減数分裂、精子形成に関わる分子機構の解明を進めた。(2)移植卵母細胞の成熟を促進するため、移植マウスにプロジェステロンチューブとFSH浸 透圧ポンプを留置したところ、体外成熟率は改善した。また、生殖細胞顕微操作の基礎検討として、卵母細胞を遠心処理して小片化した後、電気融合により再構成した卵子は単為発生能を持つことを明らかにし、雌生殖細胞の分化・成熟機構の解明を進めた。(3)これまで同定したES細胞分化関連遺伝子のうち、Ppet21遺伝子は細胞の増殖よりも分化制御に関わること、その発現はOct3/4やLIF-STAT3シグナル伝達系の直接的な支配下にないことを明らかにした。また、グロビン遺伝子の発現制御下でレポーター遺伝子が発現するベクターを開発し、細胞のエピジェネティックな差違をモニターする系を確立し、幹細胞の分化制御機構の解明を進めるとともに新しいアプローチを確立した。(4)伸長期のウシ胚における遺伝子発現解析等を行うために、試験管内で胚を伸長させる培養系を検討したところ体外受精17日間の培養で15mmまで伸長させることができた。また、無細胞発現系によるBMP等の組換えタンパク質の生産系を確立し、着床・胎盤形成機序と機構を解明の基盤を確立した。
